5-HT7 receptor stimulation and blockade: a therapeutic paradox about memory formation and amnesia

چکیده
OVERVIEW Mammalian memory involves multiple brain areas and neurotransmitter systems; both at the receptor and post-receptor level (Meneses, 2014a,b). Investigations of serotonin (5-hydroxytryptamine, 5-HT) involvement in memory (Meneses, 2013) have been significantly enhanced by the identification, classification, and cloning of multiple receptors (Hoyer et al., 1994, 2002) and studies at the post-receptor level (Raymond et al., 2001). Emergent investigation of 5-HT7 receptor is growing but contradictory. Hence, major current questions include (1) the paradox that 5-HT7 receptor agonists and antagonists in behavioral memory tasks are revealing promnesic and antiamnesic effects; (2) 5-HT7 and 5-HT1A receptors interaction (and other 5-HT receptors), and neural markers associated to these cognitive processes and individual differences. In this work recent data are briefly revised.

Early and late consolidation and reconsolidation of memory in the prelimbic cortex
Rats can learn to forage among olfactory cues to associate one with reward in only 3 massed trials. The learning is achieved in less than 10 min and results in a memory trace lasting at least 1wk week. To study the neuro-anatomical circuits involved in the memory formation we used immunoreactivity to the immediate early gene c-fos as a marker for neuronal activity induced by the learning. The p...

متن کاملThe Effect of 5HT3 Agonist Receptor of the Lateral Hippocampal on Amnesia Induce by Morphine in Mice
Background and Objective: It has been shown that the opioidergic system exerts widespread effects on cognitive functions. Interactions between opioid and serotonin receptors have been reported in some brain structures such as the dorsal hippocampus, thus a functional interaction between opioids and serotonin seems possible concerning memory control. The purpose of this study was to assess the e...
متن کاملStress- and antidepressant treatment-induced modifications of 5-HT7 receptor functions in the rat brain
This paper summarizes a series of electrophysiological studies aimed at finding the effects of the activation of 5-HT7 receptors on neuronal excitability as well as on excitatory and inhibitory synaptic transmission in the hippocampus and in the frontal cortex of the rat. These studies demonstrated that 5-HT7 receptors play an important role in the modulation of the activity of the hippocampal ...
متن کاملEffects of the Selective 5-HT7 Receptor Antagonist SB-269970 and Amisulpride on Ketamine-Induced Schizophrenia-like Deficits in Rats
A wide body of evidence suggests that 5-HT7 receptors are implicated in a variety of central nervous system functions, including control of learning and memory processes. According to recent preclinical data, the selective blockade of these receptors may be a potential target for cognitive improvement in schizophrenia. The first aim of the present study was to evaluate the effects of the select...
